Technology Background
1.) Three-Plasmid System (2nd Generation)
The most common method for lentivirus production, this system consists of:
- Transfer Plasmid
- Packaging Plasmid (Gag, Pol, and Rev integrated into a single vector)
- Envelope Plasmid

2. Four-Plasmid System (3rd Generation)
This system consists of:
- Transfer Plasmid
- Two Packaging Plasmids (one encoding Gag/Pol and another encoding Rev)
- Envelope Plasmid
While the 3rd-generation system enhances safety compared to the 2nd generation, it generally produces lower viral titers. By default, we use the 3rd-generation system, unless otherwise requested by the customer.

- Lenti-Selection Marker Constructs: Includes Puromycin, Blasticidin (BSD), Neomycin, etc., for stable cell line screening.
