Overview

Lentivirus packaging services provide researchers with high-efficiency lentiviral vectors for gene delivery into a wide range of target cells, including both dividing and non-dividing cells. Lentiviral vectors, derived from the Human Immunodeficiency Virus (HIV), are widely utilized in gene therapy, functional genomics, stable cell line development, and various research fields, including high-throughput gene function screening and transgenic animal model development.

Technology Background

1.) Three-Plasmid System (2nd Generation)

The most common method for lentivirus production, this system consists of:

  • Transfer Plasmid
  • Packaging Plasmid (Gag, Pol, and Rev integrated into a single vector)
  • Envelope Plasmid

2. Four-Plasmid System (3rd Generation)

This system consists of:

  • Transfer Plasmid
  • Two Packaging Plasmids (one encoding Gag/Pol and another encoding Rev)
  • Envelope Plasmid

While the 3rd-generation system enhances safety compared to the 2nd generation, it generally produces lower viral titers. By default, we use the 3rd-generation system, unless otherwise requested by the customer.

2. Lentivirus Subcloning

To enhance functional detection, we offer gene subcloning into the following constructs:

  • Lenti-mini GFP construct (~330bp): A compact mini-GFP tag used for rapid expression analysis post-transduction.
  • Lenti-Selection Marker Constructs: Includes Puromycin, Blasticidin (BSD), Neomycin, etc., for stable cell line screening.

 

3. Lentivirus Packaging

  • Production of high-titer lentiviral stocks using optimized protocols for maximum yield and purity
  • Comprehensive quality control for titer, purity, and functionality

 

4. Functional Testing

Available assays based on construct type and project requirements, including:

5. Advanced Endotoxin Removal

  • Our plasmids are endotoxin-free, significantly reducing endotoxin levels in viral preparations
